SPECIFICITY OF ACTION OF 3 POSSIBLE ANTAGONISTS OF AMINO ACID-INDUCED NEURONAL EXCITATIONS

The actions of 3 compounds, glutamic acid diethylester, 1-hydroxy-3-aminopyrrolidone-2 and L-5,6-dimethoxyaporphine (nuciferine) which have been claimed to be antagonists of excitatory amino acid and particularly of glutamate excitations of central neurons, were compared in rats. Suitable electrophoretic doses of glutamic acid diethylester antagonized the effects of glutamate and aspartate to a much greater extent than those of homocysteate or acetylcholine. 1-Hydroxy-3-aminopyrrolidone-2 distinguished between glutamate/aspartate and homocysteate but not acetylcholine. Nuciferine showed no selectivity amongst the amino acids. Large applications of any of the compounds would antagonize the effects of all of the excitatory agonists.
