颈动脉注射甘露醇对血脑屏障通透性及甲氨喋呤入脑影响的实验研究

摘要：

本文用25％甘露醇注入兔的一侧颈内动脉,获得了满意的渗透性BBB开放模型,使静脉注射的EB在BBB开放侧脑半球着色。这种BBB开放是可逆性的,约持续2小时,且不伴神经组织结构的损害。在BBB开放后的1、24、72小时,脑组织水分、Na+Kt含量均在正常范围,与对照组比较无显著性差异（P>0.05）。超微结构观察的结果证实,渗透性BBB开放的形态学基础主要是脑毛细血管内皮细胞的皱缩,其间的紧密连接增宽。在BBB开放期间,由颈动脉或静脉应用MTX,能显著提高脑组织内的MTX浓度,前一途径可使BBB开放侧脑半球的药物浓度增至最高水平。本文研究结果提示:渗透性BBB开放是一种安全、简单可行的方法,为化疗药物入脑开辟了途径,有可能改善恶性脑瘤的化疗效果,对其它颅内疾病的治疗及某些基础学科的研究也具有意义。
