重组人促红细胞生成素对急性脑损伤大鼠的神经保护作用研究

摘要
目的探讨重组人促红细胞生成素(rh-EPO)对大鼠急性脑损伤后脑细胞凋亡的保护作用及对GLT-1/GLAST表达的影响。方法 60只大鼠分为对照组(n=18)、急性脑损伤组(n=22)和rh-EPO处理组(n=20),急性脑损伤组用改良Feeney′s自由落体打击器制作急性脑损伤模型,rh-EPO处理组采用同样的方法制作脑外伤模型15min后腹腔注射rh-EPO,所有动物在实验完成48h后断头取脑。应用RT-PCR、Western blot检测GLT-1、GLAST mRNA及蛋白表达。结果急性脑损伤48h后,大鼠缺血区GLT-1和GLAST mRNA及蛋白表达较对照组均显著降低(均P<0.01),而rh-EPO处理组GLT-1和GLAST的mRNA及蛋白表达较急性脑损伤组均显著增加(均P<0.01)。结论 EPO可显著减轻大鼠急性脑损伤,其机制可能与GLT-1/GLAST表达上调有关。
