摘要
生长相关蛋白43(GAP-43)是一种细胞内生长相关蛋白,在成人中与突触可塑性相关。在第二信使信号通路中,GAP-43参与信号传递过程。GAP-43的Ser-41是蛋白激酶C的磷酸化位点,当神经营养因子与GAP-43结合激活GTPases酶使GAP-43磷酸化,磷酸化的GAP-43可促进伪足生长、轴突分支和肌动蛋白在细胞膜的聚集。另外,GAP-43的磷酸化也破坏了GAP-43与钙调素的结合,释放的GAP-43可以作为肌动蛋白纤维和细胞膜的桥梁。GAP-43的N末端存在与G蛋白作用的结构域,介导GAP-43与质膜和脂伐的结合。GAP-43与突触可塑性有关。以前认为,GAP-43只存在于已分化定型、有轴突生长的成熟神经元中。现在研究发现,GAP-43在神经前体细胞(NPCs)中就有表达。体内、体外的实验结果均显示,GAP-43去表达后神经分化会被抑制,说明GAP-43在神经发生过程中参与决定细胞命运。本文着重介绍近年来国内外对GAP-43的分子机制及生理作用等方面的新认识。
