星形胶质细胞调节慢性疼痛的分子机制

摘要
慢性疼痛是临床治疗上的难题之一,开发新的镇痛药物需要对慢性疼痛的产生和维持机制进行更加深入的研究。近年来的研究表明,神经胶质细胞,尤其是中枢神经系统的小胶质细胞和星形胶质细胞在慢性疼痛的发生发展中起重要作用。星形胶质细胞是中枢神经系统中数量最多、占据体积最大的细胞,在结构上与神经元突触紧密接触。而且,外周神经损伤、组织损伤、肿瘤浸润、关节炎等引起的慢性疼痛状态下,脊髓星形胶质细胞比小胶质细胞呈现出更持久的活化,该活化与慢性疼痛行为密切相关。活化的星形胶质细胞可通过丝裂原激活的蛋白激酶/MAP激酶(MAPK)释放炎症前细胞因子、趋化因子等调节神经元的兴奋性,促进慢性疼痛的维持。本文对脊髓星形胶质细胞参与慢性疼痛调节的机制研究进展进行了综述。
