<正>1 细胞凋亡研究简史细胞凋亡的研究可以上溯至上世纪中叶,但其在机体发育和细胞内稳态上的作用只是在近十年来才被生物学界所认识.早在1858年,Virchow就发现正常组织中存在着持续的,有别于病理坏死的一种细胞死亡.这一发现导致了随后一个世纪中对凋亡的形态学尤其是染色质的变化详细的研究观察.Graper于1914年提出细胞凋亡和细胞分裂相辅相成,是体内维持细胞自稳态的重要机制.De Duve随后证明凋亡是一个主动过程.同时,许多研究证明了细胞凋亡是胚胎发育或变态中一个必不可少的过程.细胞凋亡(apoptosis)这一定义是Kerr和同事于1972年提出的.随后在1980年,Wyllie报道皮质激素引起的胸腺细胞凋亡伴有特征性的染色体梯度断裂.这一发现为凋亡研究提供了实验基础.在随后的数年内,与凋亡有关的基因相继被发现,参与诱发或抑制凋亡的生长因子陆续被报道.凋亡的生物化学过程逐步被阐明.
