XRCC1与脑缺血再灌注DNA单链断裂

摘要
目的 观察脑缺血再灌注损伤中XRCC1表达、DNA单链断裂、神经元凋亡的变化 ,探索它们的关系。方法 应用线栓法建立大鼠局灶性脑缺血再灌注模型 :免疫组化检测XRCC1蛋白表达 :单细胞凝胶电泳检测细胞DNA单链断裂 :流式细胞检测细胞凋亡。结果 XRCC1在缺血再灌注 10min后即出现降低 ,1h后整个大脑中动脉供血皮质和基底节区XRCC1进一步下降 ,一直持续到 4 8h。单细胞凝胶电泳示缺血再灌注 10min基底节区出现DNA单链断裂 ,12h达到高峰 ,4 8h仍有DNA单链断裂。流式细胞显示缺血再灌注 4h出现大量细胞凋亡 ,2 4h达到高峰 ,4 8h仍保持较高的水平。结论 在脑缺血再灌注中 ,XRCCl早期降低是DNA单链断裂无法修复的机制之一 ;大量DNA单链断裂和细胞凋亡相关。
