髓样分化因子88抑制剂和氧化低密度脂蛋白对单核巨噬细胞基质金属蛋白酶-8表达的影响

摘要
目的观察氧化低密度脂蛋白(oxLDL)和髓样分化因子88(MYD88)抑制剂对小鼠单核巨噬细胞RAW264.7炎症基因表达的影响。方法采用不同剂量的oxLDL刺激RAW264.7细胞,并用不同剂量MYD88抑制剂处理oxLDL 75μg/ml组,应用ELISA和逆转录-聚合酶链反应检测基质金属蛋白酶-8(MMP-8)及其mRNA表达的变化。结果 RAW264.7细胞经oxLDL刺激后MMP-8表达明显升高,且呈时间和剂量依赖性。MYD88抑制剂可抑制oxLDL刺激引起的RAW264.7细胞MMP-8表达升高。结论 MYD88抑制剂可逆转oxLDL诱导的RAW264.7细胞MMP-8高表达。
