Activation of neocortical 5 hydroxytryptamine 2A (5-HT 2A ) receptors is thought to mediate the profoud psychominetic effects of hallucinogenic drugs such as lysergic acid diethylamide(LSD).Coversely, blockade of neocortical 5-HT 2A receptor may be related to the thymoleptic effects of newly released antidepressant especially atypical antipsychotic drugs. Electrophsiological experiments using in vitro rat slices of the medial prefrontal cortex has found that activation of 5-HT 2A receptors results in glutamate release from thalamocortical terminals.In addition to activation of 5-HT 2A receptors, metabotropic glutamate (mGluR),and neuropeptide (μ-opioid) receptors suppress this release of glutamate that is induced by 5-HT 2A receptor activation. Recent clinical reports have found clear structural change in the prefrontal cortex of depressed patients. Furthermore, a number of post-mortem studies of suicides from different laboratories have found an increased number of 5-HT 2A binding sites in certain regions of the prefrontal cortex. Thus,understanding the effects of 5-HT 2A receptor activation in prefrontal cortex is likely to be relevant for the development of antidepressant drugs.
