基于代谢组学的补阳还五汤干预缺血性脑卒中的作用机制研究

摘要
目的:利用代谢组学技术探讨补阳还五汤干预缺血性脑中风的作用机制。方法:采用线栓法制备大鼠中动脉局灶性脑缺血模型(MCAO),采用神经功能积分和脑梗死体积测定方法验证补阳还五汤干预缺血性脑中风的药效;采用UPLC-MS测定假手术组、模型组和补阳还五汤给药组大鼠血清中的内源性物质的峰面积,采用偏最小二乘法(Partial Least Squares,PLS-DA)和主成分分析法(Principal Component Analysis,PCA)统计方法进行数据模式分析。结果:与模型组比较,补阳还五汤组神经功能积分显著下降(P<0.05),半脑梗死面积显著降低(P<0.05);PCA散点图中,假手术组、模型组和补阳还五汤组分离良好;内源性代谢物质谷氨酰胺、异亮氨酸、次黄嘌呤、前列腺素2、谷氨酸、脯氨酸、焦谷氨酸、缬氨酸、胆固醇和吲哚丙酸在三组中变化显著(P<0.05~0.01),为缺血性脑中风的潜在生物标志物。结论:补阳还五汤对局灶性脑缺血模型有明显的神经保护作用,作用机制可能和调节内源性代谢物变化有关。
