乌梅丸通过促进肿瘤相关巨噬细胞M1极化抗转移性结直肠癌的机制

摘要
目的:探索乌梅丸对转移性结直肠癌中肝转移和肺转移这一进程的影响及其潜在的作用机制。方法:首先随机将小鼠分为空白组,乌梅丸低、高剂量组(20、40 g·kg-1)和紫杉醇组(0.01 g·kg-1),然后建立小鼠结直肠癌肝转移和肺转移模型。给药干预4周,记录小鼠体质量,检测转移性结直肠癌小鼠的肝脏、肺脏质量和生存期。采用苏木素-伊红(HE)染色检测病理明确乌梅丸对肠癌小鼠肝、肺转移的影响。然后采用实时荧光定量聚合酶链式反应(Real-time PCR)检测乌梅丸对小鼠肠癌肝转移肝组织及肠癌肺转移肺脏组织中M1型和M2型巨噬细胞标志物mRNA水平的影响。最后通过流式细胞术检测乌梅丸对小鼠肠癌肝转移肝组织及肠癌肺转移肺脏组织中M1型巨噬细胞标记物CD80、分化抗原86(CD86)含量的影响。结果:在结直肠癌肝/肺转移小鼠模型中,与空白组比较,乌梅丸高、低剂量组和紫杉醇组肠癌肝/肺转移小鼠体质量、肝/肺质量显著降低(P<0.01),肝/肺转移小鼠的生存期显著延长(P<0.01)。组织病理学结果显示,与空白组比较,乌梅丸抑制了肠癌肝、肺转移灶的转移。Real-time PCR结果显示与空白组比较,乌梅丸能够显著升高结直肠癌肝、肺转移小鼠肝、肺组织中M1型巨噬细胞相关标记物的白细胞介素(IL)-1β、IL-6、肿瘤坏死因子-α(TNF-α)、诱导型一氧化氮合酶(iNOS)、前列腺素内过氧化物合酶2(PTGS2) mRNA水平(P<0.01),同时降低M2型巨噬细胞相关标记物精氨酸酶1(Arg1)、CD163和甘露糖受体(CD206)mRNA水平(P<0.01)。同时,流式结果表明,与空白组比较,乌梅丸显著增加M1型肿瘤相关巨噬细胞标记物CD86和CD80的含量(P<0.01)。此外,免疫组化结果显示乌梅丸促进肝转移、肺转移小鼠肝脏、肺脏组织中CD86的表达增加,抑制CD206的表达。结论:乌梅丸能够通过促进肠癌肝、肺转移中M1型肿瘤相关巨噬细胞极化来抑制结直肠癌的肝、肺转移。
