基于燕麦的抗氧化活性肽的虚拟筛选研究

摘要
Keap1-Nrf2通路在机体氧化应激损伤中发挥着关键的防御作用,对Keap1蛋白的抑制作用可以激活体内的Nrf2蛋白,具有促进抗氧化基因表达和清除ROS的作用.燕麦中存在大量的多糖和多肽分子,其中燕麦多肽具有抗氧化等多种生物活性功能.本文从文献中调研了从燕麦中提取鉴定得到的多肽,构建了包含199个燕麦多肽的数据库,并基于Keap1蛋白对多肽数据库进行了虚拟筛选.针对筛选获得的4个多肽分别进行了300 ns的分子动力学模拟.从模拟之后稳定结合的构象分析了不同多肽分子与Keap1的相互作用信息,并分别计算了不同多肽分子与Keap1的结合自由能.结果表明,虚拟筛选得到的多肽分子均可以与Keap1蛋白稳定结合,其中多肽27与Keap1的亲和力相对较强,可以作为后续Keap1抑制剂设计和改造的先导分子.模拟结果对于更深入理解Keap1的结构特点以及与燕麦多肽的分子识别机制具有指导意义.
